IDE, TAKEHIKO,JURGEN STEINKE, AND GEORGE F. CAHILL, JR. Metabolic interactions of glucose, lactate, and /I-hydroxybutyrate in rat brain slices. Am. J. Physiol. 217(3) : 784-792. 1969.-The metabolism of glucose, lactate, and P-hydroxybutyrate was studied in rat brain slices by following the incorporation of 14C into COz, total lipids, fatty acids, and glycogen singly or in the presence of other unlabeled substrate. Glucose-14C incorporation into COZ, lipids, and glycogen was proportional to glucose concentration in the incubation medium; however, the concentration needed for saturation of these three pathways differed. The activity of the direct oxidative shunt was negligible. Lactate and P-hydroxybutyrate carbon were both incorporated into total lipid and fatty acid proportional to their concentration. In contrast to glucose, neither lactate nor fl-hydroxybutyrate carbon was incorporated into glycogen. However, the addition of glucose to the medium induced a significant incorporation of lactate carbon into glycogen. Addition of lactate or @-hydroxybutyrate inhibited the incorporation of glucose carbon into CO 2, total lipid, and fatty acid, but stimulated its incorporation into glycogen. Both glucose and lactate markedly enhanced the metabolism of P-hydroxybutyrate to CO2 and lipid, suggesting a permissive action of glucose on P-hydroxybutyrate utilization.

Glucagon stimulates ketone utilization by rat brain slices.

Glucagon has been shown previously to increase further the enhanced tolerance for hypoxia observed in mice with elevated blood ketones. Glucagon is also known to increase blood glucose and to alter directly the metabolism of some (liver) cells. Both the increase in blood glucose and altered cellular metabolism could contribute to the increase in tolerance for hypoxia observed in mice given gluc...

Acetoacetate metabolism in infant and adult rat brain in vitro.

1. Acetoacetate or dl-beta-hydroxybutyrate increases the rate of oxygen consumption to a smaller extent than that brought about by glucose or pyruvate in adult rat brain-cortex slices but to the same extent as that in infant rat brain-cortex slices. 2. The rate of (14)CO(2) evolution from [1-(14)C]glucose considerably exceeds that from [6-(14)C]glucose in respiring infant rat brain-cortex slice...

Effects of fatty acids, ketone bodies, lactate and pyruvate on glucose utilization by guinea-pig cerebral cortex slices.

1. Starvation did not affect the rates of glucose utilization or lactate formation by guinea-pig cerebral cortex slices. 2. Palmitate (1mm), butyrate (5mm) or acetoacetate (5mm) did not affect glucose utilization or lactate formation by cerebral cortex slices from guinea pigs starved for 48hr. 3. dl-beta-Hydroxybutyrate (10mm) increased the formation of lactate without affecting glucose utiliza...

The fate of lactate in isolated cells from early neonatal rat brain. Comparison with glucose and 3-hydroxybutyrate.

Different studies suggest that lactate can be used as a fuel to meet the energy requirements of the developing brain. Lactate has been proved to be not only an energetic substrate but and important iipogenic precursor Ill. However. the use of brain slices for the study of metabolism has some limitations. such as the diffusion of substrates and oxygen to the different layers of cells. Thus, the ...

The metabolic effects of estriol in female rat liver.

The effects of estriol on oxygen uptake, glucose release, lactate and pyruvate production, beta-hydroxybutyrate and acetoacetate production in perfused rat liver as well as, carbon uptake in rat liver and intracellular calcium in isolated Kupffer cells were investigated. Basal oxygen consumption of perfused liver increased significantly in estriol or ethanol-treated rats. But these increased ef...
